Crystallization and preliminary X-ray diffraction studies of infectious bronchitis virus nonstructural protein 9.

چکیده
Avian infectious bronchitis virus (IBV), which causes respiratory disease in infected birds, belongs to coronavirus group 3. IBV encodes 15 nonstructural proteins (nsp2-nsp16) which play crucial roles in RNA transcription and genome replication. Nonstructural protein 9 (nsp9) has been identified as a protein that is essential to viral replication because of its single-stranded RNA-binding ability. The gene segment encoding IBV nsp9 has been cloned and expressed in Escherichia coli. The protein has been crystallized and the crystals diffracted X-rays to 2.44 A resolution. They belonged to the cubic space group I432, with unit-cell parameters a = b = c = 123.4 A, alpha = beta = gamma = 90 degrees . The asymmetric unit appeared to contain one molecule, with a solvent content of 62% (V(M) = 3.26 A(3) Da(-1)).

Purification, crystallization and preliminary X-ray analysis of nonstructural protein 2 (nsp2) from avian infectious bronchitis virus.
Avian infectious bronchitis virus (IBV) is a member of the group III coronaviruses, which differ from the other groups of coronaviruses in that they do not encode the essential pathogenic factor nonstructural protein 1 (nsp1) and instead start with nsp2. IBV nsp2 is one of the first replicase proteins to be translated and processed in the viral life cycle; however, it has an entirely unknown fu...

Infectious bronchitis virus (IBV) is the prototype of the genus Coronavirus. It causes a highly contagious disease which affects the respiratory, reproductive, neurological and renal systems of chickens, resulting great economic losses in the poultry industry worldwide. Avian infectious bronchitis virus (IBV) encodes 15 nonstructural proteins (nsps) which play crucial roles in RNA transcription and genome replication. One of them, nsp3, contains an ADRP (adenosine diphosphate-ribose-1'-phosphatase) domain which was revealed in recent studies to have ADP-ribose-1'-monophosphatase (Appr-1'-pase) activity. Appr-1'-pase catalyzes the conversion of ADP-ribose-1'-mo...
متن کاملMolecular Cloning and Expression of Nucleocapsid Gene of Chicken Infectious Bronchitis Virus Strain Massachusetts H120
Chicken infectious bronchitis virus (IBV) causes severe respiratory and renal dysfunction in chickens. In the present study, we used reverse transcription-polymerase chain reaction (RT-PCR) to amplify the nucleocapsid (N) protein gene of strain Massachusetts (Mass) H120 of IBV that is commonly used in the vaccine production in Iran. The PCR product with the expected size of 1.2 kb was cloned in...
متن کاملCrystallization and preliminary X-ray diffraction studies of a surface mutant of the middle domain of PB2 from human influenza A (H1N1) virus.
In the last hundred years, four influenza pandemics have been experienced, beginning with that in Spain in 1918. Influenza A virus causes severe pneumonia and its RNA polymerase is an important target for drug design. The influenza A (H1N1) virus has eight ribonucleoprotein complexes, which are composed of viral RNA, RNA polymerases and nucleoproteins. PB2 forms part of the RNA polymerase compl...
